705 Particulate matter induces inflammatory response in human outer root sheath cells via oxidative stress-dependent MAPK and JAK-STAT signaling pathways

Particulate matter (PM), a major air pollutant has been demonstrated to cause intracellular inflammation by inducing reactive oxygen species (ROS) generation in human keratinocytes. Although PM shown be associated with various skin disorders, including atopic dermatitis, eczema, and aging, there have few studies on the effect of hairs. The aim this study was investigate potential role oxidative stress-dependent MAPK JAK-STAT signaling pathways outer root sheath (ORS) cells induced PM. ORS were treated at doses for 3 24 hours. Intracellular ROS detected using chemical fluorescent probe fluorescence plate reader. Gene protein expressions aryl hydrocarbon receptor (AhR), p38, JNK, JAK1, JAK2, JAK3, STAT1, STAT3, STAT5, cyclooxygenase 2 (COX-2) determined reverse transcription-polymerase chain reaction western blotting, respectively. levels prostaglandin E2 (PGE2) proinflammatory cytokine expression, that matrix metalloproteinases (MMPs) interleukins (ILs) analyzed enzyme-linked immunosorbent assay. increased via activation AhR. also STAT3 level inflammatory mediators cytokines COX-2, PGE2, IL-1α, IL-1β, IL-6, IL-8, MMP-1. Pretreatment N-acetyl-L-cysteine attenuated increase cytokines. Also, pretreatment inhibitors decreased stimulated These results suggest can response which might impair hair growth.
